Research on the biomass process of Taxus wallichiana Zucc. callus and solutions to improve Taxane content in biomass.

Result:

  • Building the process of biomass Taxus wallichiana Zucc. Callus: Inoculation time to limit browning of samples is 15 days. Culture medium should be pretreated with Acid ascorbic 2% and AgNO3 150 ppm to limit sample browning and stimulate callus formation and increase biomass.
  • Provide a solution to improve the content of Paclitaxel in callus:
  • State 1: Stimulation of callus growth with B5 medium supplemented with 0.1ppm 2,4-D; 0,5ppm 6-BA and 0,5ppm NAA and 150 ppm AgNO3.
  • State 2: Stimulation of Paclitaxel compound accumulation in callus by inoculation of callus on B5 medium supplemented with 0.1ppm 2,4-D; 0,5ppm 6-BA and 0,5ppm NAA and 150 ppm AgNO3 and Methyl jasmonate 5ppm, VOSO4 0,2mM or Methyl jasmonate 5ppm, Salicylic acid 50ppm and VOSO4 0,2mM.
