Building one process of biomass Taxus wallichiana Callus by bioreactor to obtain paclitaxel for cancer treatment includes steps:
- State 1: Multiply biomass in an erlen 250ml: mass cell/medium volume with rate 1/10 cultured in erlen 250ml containing medium B5, 20 /l saccharose, 5 g/l glucose, 5 g/l fructose, 150 mg/l AgNO3, 1 mg/l picloram, 0,1 mg/l kinetin, 0,5 mg/l NAA, at dark conditions, swing at speed 60 rpm, temperature 25±1oC. After 2 weeks, 20 g/l fructose was added to the culture medium and continued under the same conditions. After 1 weeks of adding sugar, harvesting Taxus cells.
- State 2: Permanent of Taxus cells in 1% calcium alginate, stand for 24 hours to solidify the newly formed Calcium alginate particles.
- State 3: After 24 hours of permanenting, Taxus cells were coated with calcium alginate 1% put in a fermenter and perform cell culture in bioreactor. Culture conditions: temperature 25±1oC, swing at speed 60 rpm, at dark. pH = 5,7; DO 84,9. Gas content: Air 89,5%, O2 10% và CO2 5%, mix with 3 gas.
- State 4: After 1 week of culture in the bioreactor, the biosynthesis inducer paclitaxel was added to the fermenter containing 5 mg/l Methyl jasmonate and 0.2 mM VOSO4.
- State 5: After 24 weeks of adding the inducers, collection of Taxus cell biomass to receive paclitaxel for cancer treatment.
